Stereology is a valuable tool for neuroscientists, allowing them to obtain 3-Dimensional information from 2-Dimensional measurements made on appropriately sampled sections. This 3-D information is invaluable in correlating structural/functional relationships in the pursuit of far greater understanding of the function of the central nervous system. However, in carrying out such measurements, often based on limited data sets, there is a risk of experimenter bias. An important feature of modern design based stereology is to be aware of potential sources of bias (...) and eliminate them during the data collection. With many of the major neuroscience journals now insisting that quantitative data be presented, there is a greater need than ever for neuroscientists to understand the theory and practice behind quantitative methods, such as those offered by stereology. Quantitative Methods in Neuroscience is a cookbook of stereological methods written especially for neuroscientists. It provides clear and accessible advice about when and when not to use stereology. In his doctoral dissertation On the Principle of Sufficient Reason, Arthur Schopenhauer there outlines a critique of Euclidean geometry on the basis of the changing nature of mathematics, and hence of demonstration, as a result of Kantian idealism. According to Schopenhauer, Euclid treats geometry synthetically, proceeding from the simple to the complex, from the known to the unknown, “synthesizing” later proofs on the basis of earlier ones. Such a method, although proving the case logically, nevertheless fails to attain the raison (...) d’être of the entity. In order to obtain this, a separate method is required, which Schopenhauer refers to as “analysis,” thus echoing a method already in practice among the early Greek geometers, with however some significant differences. In this essay, I here discuss Schopenhauer’s criticism of synthesis in Euclid’s Elements, and the nature and relevance of his own method of analysis. (shrink)

Following the Kantian critique of metaphysics, the conscious subject is discovered to be an insurmountable obstacle with respect to knowledge of things themselves. For this reason, Kant concludes that metaphysics as the science of being as being is impossible. In this essay, the possibilities of metaphysics in light of the problem of subjectivity are reexamined. The nature and relationship between the conscious subject and the embodiment of the subject is first examined. Following this, the subject’s “encounter with being” within consciousness (...) is analyzed yielding two fundamental structures that are determined to hold with respect to being in relation to the conscious subject. Further examination of the act of reflection coupled with judgment reveals that these structures may in fact be transcended, from which an encounter with being as such follows. On the basis of reflection and judgment, metaphysics is thereby determined to be possible. (shrink)

Aims and background: The present work assessed the impact of two decrees on ethics committees in Italy, aimed at bringing the national laws on the conduct of clinical trials into line with the rest of the EC, and regulating and facilitating not-for-profit research.Material and methods: Prospectively collected data from an Italian multicentre study were examined with respect to the ethics review process. Administrative and time elements of the review process were audited. Main outcome measures were time between the application submission (...) and the ethics committee definitive opinion, type and number of application submission forms, number of ethics committees that refused fee exemption, and time between the ethics committee approval and the administrative authorisation.Results: A total of 134 local research ethics committees were approached. Application submission procedures and application forms varied greatly; paper submission was mandatory. The median time from submission to approval was 72 days. Only two LRECs refused the fee exemption. The median time from LREC approval to administrative agreement was 50 days and only 9.6% of local authorities came to a verbal agreement with the sponsor.Conclusions: Italian LRECs are still not sufficiently efficient in complying with the Directive 2001/20/EC requirement . Better coordination of LRECs work is needed although the optimal level of coordination between them is still not known. In the meantime, national guidelines are needed concerning the application of Directive 2001/20/EC. The behaviour of Italian LRECs towards not-for-profit research was excellent although only the fee exemption was requested. (shrink)

During early embryonic development in several metazoans, accurate DNA replication is ensured by high number of replication origins. This guarantees rapid genome duplication coordinated with fast cell divisions. In Xenopus laevis embryos this program switches to one with a lower number of origins at a developmental stage known as mid‐blastula transition (MBT) when cell cycle length increases and gene transcription starts. Consistent with this regulation, somatic nuclei replicate poorly when transferred to eggs, suggesting the existence of an epigenetic memory suppressing (...) replication assembly origins at all available sites. Recently, it was shown that histone H1 imposes a non‐permissive chromatin configuration preventing replication origin assembly on somatic nuclei. This somatic state can be erased by SSRP1, a subunit of the FACT complex. Here, we further develop the hypothesis that this novel form of epigenetic memory might impact on different areas of vertebrate biology going from nuclear reprogramming to cancer development. The aim of this essay is to show that phenomenal properties are contentless modes of appearances of representational properties. The essay initiates with examination of the first-person perspective of the conscious observer according to which a “reference to I” with respect to the observation of experience is determined. A distinction is then drawn between the conscious observer and experience as observed, according to which, three distinct modifications of experience are delineated. These modifications are then analyzed with respect to the content (...) of experience and from this the ground of the distinction between phenomenal and representational properties is identified. Sleep is a major concern, especially in people with Down Syndrome. Beyond Obstructive Sleep Apnea, a number of other sleep difficulties have been reported in children with DS, such as delayed sleep onset, night-time awakenings, and early morning awakenings. The detrimental effect of sleep difficulties seems to contribute to and exacerbate the cognitive and behavioral outcomes of DS. Although the screening for sleep disorders is recommended early in age in DS, only a few studies have evaluated the sleep profile in (...) preschool-age children with DS. The aim of the current study was to assess the association between sleep disturbances and behavioral problems in a group of preschool-age children with DS, by means of a feasible and easy-to-administer parent-report questionnaires. Seventy-one preschool-age children with DS, ranging in age from 3 to 5.11 years, were included in this retrospective study. Sleep disturbances were evaluated by means of the Sleep Disturbance Scale for Children, while emotional and behavioral problems by means of the Child Behavior Checklist. Sleep breathing disorders were the most frequent sleep difficulties reported by parents. Moreover, children with clinical scores in total sleep problems exhibited elevation of psychopathological symptoms, namely Total problems, Affective problems, Anxiety problems, Pervasive Developmental Problems, and Attention Deficit/Hyperactivity Problems. The identification of the broader connection between sleep difficulties and emotional and behavioral problems in preschool-age children with DS leads to important considerations for intervention. BackgroundPositive psychosocial factors can play an important role in the development of cardiovascular disease. Among them, psychological resilience is defined as the capacity of responding positively to stressful events. Our aim was to assess whether PR is associated with CVD or metabolic disturbances through a systematic review.MethodsWe gathered articles from PubMed, Web of Science, PsycInfo, and Google Scholar up to October 28, 2021. We included articles that were in English, were observational, and had PR examined as exposure. The CVD outcomes (...) were either clinical or metabolic outcomes.ResultsOur literature search identified 3,800 studies, of which 17 met the inclusion criteria. Of them, seven were longitudinal and 10 cross-sectional, and 13 were on adults and four on children. The exposure assessment was heterogeneous, i.e., 12 studies used different kinds of self-administered questionnaires and five used interviews with a psychologist. Regarding outcomes, five studies investigated CVD, seven obesity, one metabolic syndrome, two hypertension, four dyslipidemia, and four diabetes. In longitudinal studies, PR was found to have an inverse association with included outcomes in five studies from the Swedish military conscription cohort but had no association with CVD in a study on African-American women and was associated with slower progression of diabetes in a general population. The cross-sectional studies showed that the prevalence of disease was not associated with PR in many cases but the progression of disease was associated with PR.ConclusionPR seems to have a possibly favorable association with CVD and metabolic disturbances that differs according to the type of outcome and population. The environment in which humans evolved is strongly and positively autocorrelated in space and time. Probabilistic judgments based on the assumption of independence may not yield evolutionarily adaptive behavior. A number of “faults” of human reasoning are not faulty under fuzzy arithmetic, a nonprobabilistic calculus of reasoning under uncertainty that may be closer to that underlying human decision making.

We analyze the transition of central banks from lenders to market makers of last resort. The adoption of unconventional monetary policies characterizes this transition. In their new role as market makers, central banks engage in the latter by extending and reinforcing interventions in other markets than the traditional bank reserves market. We then explain that the difference between the two roles is one of degree rather than kind. In both cases, the prevention of liquidity shortages is a primary concern. As (...) conventional policies become inadequate, central banks resort to unconventional policies to escape a general liquidity shortage at the zero lower bound. However, these unconventional policies do not solve the structural problems in financial and real markets. Both conventional and unconventional monetary policies cause price distortions, in particular on asset markets. The policies of the market maker of last resort prevent necessary readjustments of cyclical divergences between real and financial markets. (shrink)

A contentious feature in the thought of Arthur Schopenhauer is his account of the Platonic Ideas. This is no doubt evidenced by the scholarly literature where various difficulties have been identified in regards to this introduction, and often varying positions maintained. Within this essay, I offer a survey of the major debates surrounding this issue. Following this, I turn to a specific question related to Schopenhauer’s claim that his own account of the Platonic Ideas is authentic to the original views (...) of Plato himself. I show that, excepting minor points of similarity, Schopenhauer’s account is quite inauthentic to that of Plato. Still more, drawing upon his distinction between abstract and intuitive knowledge, I offer an interpretation of the relationship between Genius and contemplation employing the concept of a “circle of signification”. Finally, I identify some difficulties in relation to Schopenhauer’s views. (shrink)

In recent times, we have seen a resurgence of interest in the application of philosophy as a therapeutic for the purpose of alleviating the existential ills of human life. Within this paper, it is argued that not only can philosophy be applied as a therapeutic, but that the very act of doing philosophy is therapeutic. The paper begins with a discussion of human nature as bound to finitude and the suffering of existence. The necessity to labor along with the need (...) for relief from labor in the form of recreation and play is then discussed. Play is thereafter distinguished from leisure, and the concept of philosophical contemplation, following Aristotle, is introduced. It is argued that the activity of contemplation results in relief from the suffering of existence, and that its exercise may in consequence be considered a kind of therapeutic. (shrink)

En este sentido, el Buen Vivir representa el principio inspirador de los movimientos indígenas que proponen un proyecto alternativo que se opone al paradigma dominante y se presenta como respetuoso de las diversidades.
